Between a half hour and 45 min. depends on how many people are in the group and how many questions are asked. It is much less crowded and more enjoyable than Monticello, in my opinion. you can tour the grounds and certain parts of the house at your leisure for as long as you like.

I have an RV too, I would suggest coming in to Poplar Forest the 'back way' through the neighborhood off of Waterlick Road. Mind the overhanging trees though! The main road is pretty narrow. There are two gravel parking areas, but no 'designated' RV parking. You might ask the folks inside the visitors center if you can park on the grass next to the parking lot, they're very nice. the house tour is about 45 minutes, depending on the guide and how many questions are asked by participants. Also tours are done on specific times so you may have to wait for the next tour. Fortunately, there is a self guided component which you can do all or part of while waiting. Total time for someone who actually reads all of the guide and displays is about 2 hours. I highly recommend tours of Monticello as well. It gives more perspective to Poplar Forest.
